3-D light switch for the brain: Device may help treat Parkinson's, epilepsy; aid understanding of consciousness

ScienceDaily (Nov. 19, 2012) ? A new tool for neuroscientists delivers a thousand pinpricks of light to a chunk of gray matter smaller than a sugar cube. The new fiber-optic device, created by biologists and engineers at the Massachusetts Institute of Technology (MIT) in Cambridge, is the first tool that can deliver precise points of light to a 3-D section of living brain tissue. The work is a step forward for a relatively new but promising technique that uses gene therapy to turn individual brain cells on and off with light.

Scientists can use the new 3-D "light switch" to better understand how the brain works. It might also be used one day to create neural prostheses that could treat conditions such as Parkinson's disease and epilepsy. The researchers describe their device in a paper published November 19 in the Optical Society's (OSA) journal Optics Letters.

The technique of manipulating neurons with light is only a few years old, but the authors estimate that thousands of scientists are already using this technology, called optogenetics, to study the brain. In optogenetics, researchers first sensitize select cells in the brain to a particular color of light. Then, by illuminating precise areas of the brain, they are able to selectively activate or deactivate the individual neurons that have been sensitized.

Ed Boyden, a synthetic biologist at MIT and co-lead researcher on the paper, is a pioneer of this emerging field, which he says offers the ability to probe connections in the brain.

"You can see neural activity in the brain that is associated with specific behaviors," Boyden says, "but is it important? Or is it a passive copy of important activity located elsewhere in the brain? There's no way to know for sure if you just watch." Optogenetics allows scientists to play a more active role in probing the brain's connections, to fire up one type of cell or deactivate another and then observe the effect on a behavior, such as quieting a seizure.

Unlike the previous, 1-D versions of this light-emitting device, the new tool delivers light to the brain in three dimensions, opening the potential to explore entire circuits within the brain. So far, the 3-D version has been tested in mice, although Boyden and colleagues have used earlier optogenetic technologies with non-human primates as well.

Targeting neurons with light

One of the advantages of optogenetics is that this technology allows scientists to focus on one particular type of neuron without affecting other types of neurons in the same area of cortex. Probes that deliver electricity to the brain can manipulate neurons, but they cannot target individual kinds of cell, Boyden says. Drugs can turn neurons on or off as well, he continues, but not on such a quick time scale or with such a high degree of control. In contrast, the new 3-D array is precise enough to activate a single kind of neuron, at a precise location, with a single beam of light.

In an earlier incarnation, Boyden's device looked like a needle-thin probe with light-emitting ports along its length; this setup allowed scientists to manipulate neurons along a single line. The new tool contains up to a hundred of these probes in a square grid, which makes the device look like a series of fine-toothed combs laid next to each other with their teeth pointing in the same direction.

Each probe is just 150 microns across -- a little thicker than a human hair, and thin enough so that the device can be implanted at any depth in the cortex without damaging it. The brain lacks pain receptors, so the implants do not cause any discomfort to the brain itself. As in the earlier model, several light-emitting ports are located along the length of each probe. Scientists can illuminate and change the color of each light port independently from the others.

Adding a third dimension to the probe's light-delivery capabilities has allowed researchers to make any pattern of light they want within the volume of a cubic centimeter of brain tissue, using a few hundred independently controllable illumination points.

"It's turning out to be a very powerful and convenient tool," says MIT professor of electrical engineering Clifton Fonstad, co-lead author of the paper.

Blue for on, yellow for off

Neurons in the brain are not naturally responsive to light, so scientists sensitize these cells with molecules called opsins, light-detecting proteins naturally found in algae and bacteria. Genes for an opsin are transferred to the neurons in a mouse's brain using gene therapy, a process in which DNA is ferried into a cell via a carrier such as a harmless virus. The carrier can be instructed to deliver the DNA package only to certain types of cells.

Different colors of light turn different flavors of opsin on -- blue might cause one opsin to activate a cell, while yellow might cause another opsin to silence it. Neurons that are sensitized with opsins gain these abilities to respond to light.

The response of an individual neuron -- whether to turn on or turn off -- depends on the type of opsin it was sensitized with, and the color of light used to illuminate it. In this way, the tool gives neuroscientists an unprecedented level of control over individual neurons in the brain.

Teams from around the world are currently using the technology developed by Boyden's group to study some of the most profound questions neuroscience tries to answer, such as how memory works, the connections between memory and emotion, and the difference between being awake and being asleep.

"I'm really excited about how the brain computes -- the ebb and flow of consciousness," Boyden says. "We know so little about the brain."

A better understanding of the brain may lead to another benefit of this technology: therapy. If a particular type of cell malfunctions in a particular disease, scientists may be able to use a modified 3-D array as a neural prosthesis that could help to treat neurological conditions. Using light to stop overactive cells from firing might alleviate the uncontrollable muscle action of Parkinson's disease. Cells that cause seizures in the brain could be quieted optically without the side effects of anti-seizure medications. Implants that correct hearing deficiencies are also being explored with this technology.

Although the new device is effective in bringing light to the brain, other challenges remain before optogenetics can be used for medical therapy, Boyden says. Scientists do not yet know for certain whether the body will detect the opsin proteins as foreign molecules and reject them. Gene therapy will also have to prove itself if neurons are to be sensitized with opsin effectively.

"It's a long road," Boyden admits.

Meanwhile, he continues, the demand for the tool is currently higher than his team can supply. Boyden says his group is excited about the possibility of commercializing the new 3-D array, as one potential route that would make the devices available as quickly as possible to the neuroscience community.

The world-record-holder Sun Yang won a third gold medal at the Asian swimming championship in Dubai, United Arab Emirates, to cap another day of Chinese dominance in the pool.

Sun, a four-time Olympic medalist, won the 1,500-meter freestyle in a championship record time of 14 minutes 44.10 seconds, far short of his world record time but enough to easily win the race.

He was among eight Chinese to take gold in nine races, including the Olympic silver medalist Lu Ying, who won the women?s 100 butterfly for her second gold. HUNTSVILLE, AL (WAFF) -

A soldier who lost both of his legs while fighting in Afghanistan returned home to Sardis City Friday morning.

Army Cavalry Scout Corey Garmon was injured when an IED detonated while he was patrolling in Afghanistan in July. The injury cost Garmon both his legs.

The 24-year-old has been recovering at Walter Reed Medical Center in Maryland, but now he is home.

Garmon arrived at Huntsville International Airport at 10 a.m. He then made his way?to?Sardis City for a homecoming event at the Sardis Senior Citizens Center.

Law enforcement and the Patriot Guard gave Corey escorts through Guntersville, Albertville, and Boaz.

Garmon is a graduate of Sardis High School.

The Albertville Chamber of Commerce prepared for?a big welcome home. Residents and business owners?lined up along Hwy 431 to cheer for the wounded warrior.

Guntersville First Baptist Church also planned for a smooth homecoming. The church built a ramp at Garmon's family's home to make sure it is wheelchair-friendly.

Louis Armstrong Airport access road will get bike lanes | NOLA.com

With the 2013 Super Bowl at the Mercedes Benz Superdome 80 days away, the New Orleans City Council added a few extra stitches to New Orleans' red carpet Thursday, approving a landscaping project at Louis Armstrong New Orleans International Airport and giving the strength of law to new rules for airport taxis.

Officials have been billing the airport as the city's gateway and the taxis there as its first ambassadors. In that vein, the City Council unanimously supported the sprucing up of the access road that snakes from Veterans Memorial Boulevard to Airline Drive. The strip will soon be replete with bike lanes and fresh shrubbery along the neutral ground, according to artists' renderings. The French Quarter-style lanterns will be repaired.

The $760,000 project will be paid for by the state and Jefferson Parish, but the City Council gets to weigh in because New Orleans owns the airport. The landscaping is expected to done in time for the big game.

The council's action on taxi regulations was little more than housekeeping, but important nonetheless if officials ever want the law behind them when they go after derelict cabs.

The city won a bitter fight in August to make its roughly 1,600 cabs uniformly modernized. If cabs want to pass inspections and work on New Orleans' roads, they now have to have working air conditioners, credit card machines, GPS devices and security cameras. The drivers must speak English and know the city, and cabs can be no older than seven years starting in 2014.

The City Council extended the same rules to any taxis wishing to shuttle passengers to and from the city-owned airport in Kenner. Those drivers can buy a $200 decal from the New Orleans Aviation Board after passing inspection.

When the council approved those rules two weeks ago, it did so as a resolution. To give them the strength of laws that could be enforced, they had to be delivered as an ordinance. That happened Tuesday with the council's unanimous vote.

The council also quietly looked to the airport's future after the Super Bowl, granting it permission Tuesday to auction off 260 dormant properties that surround its runways. The airport bought the properties almost 20 years ago after Kenner won a federal lawsuit about jet noise. They've since lain fallow save for a few holdout residents who refused to sell their homes in the 1990s.

Kenner Chief Administrative Officer Mike Quigley said officials are working to rezone the properties from residential to commercial to put them back on Kenner's tax rolls. New Orleans will get the money from the initial sales, but will have to repay the Federal Aviation Administration for the seed money it contributed to buy the properties in the first place.

Selling the lots will take some planning, Quigley said. Kenner is working with UNO to devise a "bundling" plan that would keep potential buyers from parsing out more valuable properties and leaving the rest. "We want to try to sell it, but the last thing you want to do is sell the most valuable pieces and have the rest of the land languish forever," Quigley said.

The New Orleans City Council approved the measure unanimously without discussion.

Bob Rae reaches out to the West on energy policy

Interim Liberal Leader Bob Rae promoted the Liberal Party as the best way to bring national leadership and sustainable development to Canada's resource wealth, especially in the oil sands, in a speech today at the Canadian Club in Toronto.

Rae touted the Liberals as a middle ground between the Conservatives, whom he implied are employing partisan rhetoric when it comes to discussing what he called "carbon pricing," and the NDP, which he said sees resource wealth as a curse, a characterization he described as "simplistic nonsense."

He said the oil sands are a "great advantage, but like anything, can be exploited for short-term gain and then squandered."

Rae suggested a number of measures that should be adopted in the development of energy resources:

  • The full participation by First Nation, Inuit and M?tis peoples.
  • Speeding up apprenticeship programs to provide skilled trades people in the energy section.
  • End subsidies on fossil fuels.
  • Government should lead discussions about an east-west pipeline from Alberta to the Maritimes.
  • A rational approach to carbon pricing as opposed to heavy regulations, with breaks for low income regions and households.
  • A clear set of policies about foreign investment in energy resources.
  • A neutral, science-based approach to the proposed Enbridge pipeline route.

Reaching out to the West

Rae emphasized that it's "wrong-headed" to pit resources against the manufacturing section, in a clear shot at NDP Leader Tom Mulcair who has lamented that the high Canadian dollar, pumped up by oil and gas revenues, is hurting the Ontario manufacturing sector. Rae suggested instead an "industrial strategy" that would see lower taxes on manufacturing.

Rae's speech Thursday is another example of the Liberal Party reaching out to the West, a region it has been all but shut out of in recent years. The party has only four MPs in the entire region, and its support in some provinces in the last election was in the single digit range. There are no Liberal MPs from Alberta.

With a sense, perhaps, that there's nowhere to go but up, the Liberal Party, as it pursues its quest for renewal, especially in its leadership contest, is much more pivoted towards the West than it has been in years.

It's a contrast from the mood of the party in 2006 during the last Liberal leadership race. In that contest involving eight candidates, none were from the West and six were from Toronto. The winner, St?phane Dion from Quebec, campaigned on a policy he called the "green shift," in effect, a carbon tax.

The present Liberal leadership has a more western slant. It's expected that Vancouver MP Joyce Murray will announce her candidacy soon. Martha Hall Findlay, a former Ontario Liberal MP, launched her leadership bid Wednesday from Calgary. Her main organizer is Calgary-based Stephen Carter who ran the successful campaigns of Calgary Mayor Naheed Nenshi and Alberta Premier Alison Redford. David Merner of Victoria is considering a leadership attempt.

Also, the day after Justin Trudeau announced his interest in the leadership, he headed to Alberta. The name Trudeau is anathema in parts of the province because of the still-simmering western resentment about the national energy policy put in place by his father three decades ago.
